In simple words, 2-tier links are backlinks build to backlinks !
For example:
- Assume "A" is your site
- and "B" links to "A"
Now the links that link to "B" are called 2-tier links !!

So you mean to say that A is my main site and B is referring to a backlink from either approved blog comment or article submission link. am i correct?
You were right on the first part but the second part was wrong.
Tier 2 are like this
- website number 1 (your website)
- Site number 2 ( website Backlinking back to your website, also called tier 1 backlink)
- Site number 3 ( this website links to site number 2, called tier 2 backlink)

You can use anything you like - all types of links work in all tiers. I often put powerful links that I don't want people to find in Tier 2. eg edu links. I know most people checking backlinks will only look at the immediate links, and if you mix things up and put them in the layers below they have no idea how you've managed to make what seem like innocuous Tier 1's so effective.

But why do you want to do tiered links ? ok , here are the reasons
1. Helps to strengthen your links
2. Helps to get your links indexed faster
3. Looks a bit more natural if done properly
4. Benefit from link flow if channelized correctly
