Sure, you can you what ever linkbuilding method you wish.
This is hard to say, but start slow and see what will develop from this, then repeat and if all is good you can continue...
Remember always use as many different seo/linkbuilding techniques as well as social signals and mimic natural internet behavior/growth of these links.

agreed you can build backlinks to any video page just the same way you would on a regular site.. if you want good results from your youtube video though, name the video and the title with your keywords in it.. if you want to target multiple keywords then upload multiple videos and do the same... it works very well and a good link building plan will get you ranked in this way but it doesn't happen in a few days, you need about 6 weeks to see rank...
