How many backlink and time I need to reach on google #1 with low competition keyword?

It depends not on "how many", it depends on competition and quality of course. You can reach a good position with a few high quality links.

For example, i have a blog with 1 article (500 words) - this article has 10 direct links, and about 50 second tire links and i have place one with my long tail keyword from over 500k search results.

With all the updates floating around nowdays, nope, its not. Why not take a good look at your keyword and see what your competition is for these keywords. Try see how many backlinks they have, and then figure out how long it will take you to make more (naturally and organically, or its all a waste of time... IMHO).

if it's really a "low competition" a good onpage optimization will do that, i've ranked many site with just doing that.

I wish ranking a site was that easy! But it is actually not that. There are various factors associated with keyword ranking not only the 'amount'. Quality of backlink is a great factor. Although 'quality' is a broad term which is related with relevancy, link attribute and many other things.
