Mine was many years ago when my top 10 ranking site for very high competitive search term was removed from google
I suspect manually by my competitors in collaboration with some google insiders.

I started bidding on scriptlance and my hardest moment was scriptlance was sold to freelancer (GAF) and doing business on GAF was difficult for me because I was yet to gain any reputation there so hardest moment but I believe something more hard can come anytime in future. You know about your present and past but you don't know about future so hardest moment can come any day.

It wasn't removed(didnt' express my self best) from google, it was manually punished and ended for mainly all terms on about 50+ page of the SERPs.
I can only speculate what happened, but around that time I got some information what was really happening among big players trough few friends of mine which at that time had very influential connections and their friends etc.. in various google headquarters (countries)...
So when my site reached 5th place and then was dancing randomly to 3rd and 4th place someone wasn't very happy about that and my site was gone in a matter of seconds... These keyword had about 750.000 - 3.000.000+ monthly searches each and I was ranking top 10 for main 3 + many sub keywords.

There wasn't any such hardest moment, but there was a frustrating moment when one of the client site got hurt by Panda.

Always believe in yourself and you will never have any hardest moment. See the hard moment as a chance to learn and improve yourself.
